There is no tab on my week 3 blog for view or edit, so I will continue it here instead.
The 'x' technique described in the previous blog was left for 24 hrs, the oxidation recorded, and then left for another 12 (36hrs total). The following pictures demonstrate the levels after this time period (the blue dots over the 'x' represent oxidatin after 24 hrs):

The motility screening is complete, and the 7 strains of interest were recorded via camera all in comparison to WT (on the left). Only 3 of the 7 displayed motility defects, indicating that possibly the other mutants are IMO without disrupting flagellum function. A few possible explanations for this is that the flagella genes were mutated but in a way which could be compnsated by other proteins, whereas the Manganese Oxidase could not compete and thus was overactive. Also, the gene could be a small part of overall function and thus the bacteria can still swim but only slightly less than the WT (which appears to be the case in the other 3).

Temperature Oxidation Screen:
All IMO strains were subjected to either 13, 27, 30 or room temperature and oxidation was measured. The pictures show the results after ~24hrs
