Week 3- More DNA, more chlorophylls…

This week we did a few different PCR’s and ran gels- the first one turned out really well but the two after it were less than perfect. We’re hoping to run a few more and get everything straightened out to begin sequencing. We made some WC media for a cultured infection experiment that will be started on Monday. Hopefully this infection experiment will show a significant difference between non-infected and infected cells. In the growth rate measurements I am taking now, we haven’t really seen any difference in the growth rates between non-infected and the infected cells.

We did one chlorophyll measurement this week, and we measured both the acidified and non-acidified chlorophyll methods. On Wednesday we went with the USGS on a boat to the Beaver Army Terminal (BAT) in order to clean out some of the instruments (LOBO, SUNA). Unfortunately the cleaning didn’t work on the LOBO and it is not collecting data correctly anymore. We collected some water while we were at Beaver, and filtered it for chlorophylls, PC/PN, DNA, zooplankton, and for the FlowCam. We also put together a bunch of graphs and data from measuring phytoplankton growth, measuring chlorophylls, and from grazer experiments for a presentation in the lab meeting that was on Friday. I’m hoping we can get the DNA cleaned up in order to start sequencing it soon!
